Objectives: IWGSC: Integrated physical and genetic map of chromosome 3A of wheat

  

1. BAC libraries, fingerprinting and FPC assembly: a). Fingerprint one additional 55,296-clone 3AS BAC library constructed using the restriction endonuclease EcoRI. This library will provide 8-10x coverage of the 3AS chromosome arm, which together with the previously constructed library will amount to 15x coverage of the chromosome arm; b.) Fingerprint two 3AL chromosome arm-specific BAC libraries of 55,296 clones each constructed using Hind III (currently available, 106 kb average insert size, 10x coverage of the arm) and EcoRI (will be available in Nov. 2008) restriction enzymes. These two libraries will provide 15x coverage of the 3AL chromosome arm.

2. Integrated genetic and physical maps: Construct high-density genetic map of chromosome 3A including 1,536 SNP loci for anchoring of BAC contigs from the 3AS and 3AL chromosome arms utilizing next-generation sequencing technologies and a high-throughput genotyping platform; generate the integrated genetic/physical map of the 3A chromosome.

3. Development of publicly available genomic resources, participatory research and training: Develop BAC pools to facilitate high-throughput library screening for positional cloning, participatory research, training and sharing of project results and resources with the wheat genetics community; establish publicly accessible database for the project data analysis, management and dissemination; and integrate informatics resources generated by the project with the resources of the GrainGenes and Gramene databases.
